When making a right or left turn from a complete stop my 2005 Escape - V6 AWD - is knocking. The knock seems to be coming from the area of the rear differential - has anyone else had this problem and know what it might be??

Sounds can bounce, giving you a false indication of where the sound is originating from. However, old, higher mileage shocks (60K+) can cause noise from worn out rubber bushings. Sometimes, a re-torque of the bolts and a visual inspection would be the way to go.

The cheaper route is to check the spare tire for air pressure. If the spare loses pressure the sidewall will be able to flex and it will 'bounce' off the bottom of the vehicle.

The noise started at around 50,000 miles as a low grinding sound
We are at 61,000 miles and is a knock, it is only noticable after everything has warmed up - about an hour of driving.
We have checked U-joints, cv joints, wheel bearings, axles, shocks,
spare tire, tightened all loose screws on the bumper, etc

Who is 'we'? You can check wheel bearings yourself. The bearings on my '99 Taurus started making noise around 55K - both sides. It started off as a very low hum and progressed into a grinding noise once all the grease had washed out of the unit. It increased with speed.

If anything else, get the axle fluids changed along with the transfer case fluid. If you find any metal particles around the bottom after draining the oil you might have blown a clutch pack (if equipped). I know a while back, the Explorer model had some axle problems. The front axle might be tight to get at, but with an elbow ratchet you should be able to do it just fine and use RTV sealant to seal the covers back on before you torque on the bolts.
